Two
novel species
of the beer-spoilage
Lactobacillus
bacteria
were isolated
from the brewing
environment.
One species,
of which LA-2
is a representative
strain, is an
obligately heterofermentative
Lactobacillus
sp. with
a strong beer-spoilage
ability. The
other species,
of which LA-6
is representative,
is a facultatively
heterofermentative
Lactobacillus
sp. with
a weak beer-spoilage
ability. The
carbohydrate
utilization
pattern, ribotype,
and DNA-DNA
hybridization
analysis revealed
that these novel
strains were
not related
at the species
level to any
Lactobacillus
spp. examined
in this study.
However, the
16S ribosomal
DNA sequence
of LA-2 was
found to exhibit
99.5% similarity
with that of
the L. collinoides
gene, indicating
that these species
are related.
We have also
developed primer
sets for the
detection of
the LA-2 and
LA-6. Keywords:
Beer-spoilage
bacteria, Novel
species, Polymerase
chain reaction,
16S rDNA.
Dos especies
nuevas de Lactobacillus
contaminantes
de cerveza fueron
aisladas del
medio ambiente
cervecero. Una
especie, de
la cual LA-2
es una cepa
representativa,
es un lactobacilo
obligadamente
heterofermentativo
con una fuerte
habilidad contaminante
de cerveza.
Las otras especies,
de la cual LA-6
es representante,
es un lactobacilo
facultativamente
heterofermentativo
con una débil
habilidad contaminante
de cerveza.
El patrón
de utilización
de carbohidratos,
el análisis
de hibridización
y ribotipo DNA-DNA
revelaron que
estas cepas
nuevas no estaban
relacionadas
al nivel de
especie para
cualquier especie
de Lactobacillus
examinada
en este estudio.
Sin embargo,
la secuencia
del DNA ribosomal
16S de LA-2
fue encontrada
a exhibir 99.5%
de similaridad
con aquella
del gen Lactobacillus
collinoides,
indicando
que estas especies
están
relacionadas.
Nosotros hemos
también
desarrollado
series de iniciadores
para la detection
del LA-2 y LA-6.
